This MAb is specific to kappa light chain of immunoglobulin and shows no cross-reaction with lambda light chain or any of the five heavy chains. In mammals, the two light chains in an antibody are always identical, with only one type of light chain, kappa or lambda. The ratio of Kappa to Lambda is 70:30. However, with the occurrence of multiple myeloma or other B-cell malignancies this ratio is disturbed. Antibody to the kappa light chain is reportedly useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. The major constituents of mucus, the viscous secretion that covers epithelial surfaces such as those in the trachea, colon, and cervix, are highly glycosylated proteins called mucins. These glycoproteins play important roles in the protection of the epithelial cells and have been implicated in epithelial renewal and differentiation. This gene encodes an integral membrane glycoprotein found on the cell surface, although secreted isoforms may exist. MUC-4 transcripts have been detected in normal respiratory epithelium and lung. MUC-4 is a very specific (100%) and sensitive (90%) marker of lung adenocarcinomas and is negative for mesotheliomas. Reportedly, MUC-4 expression in invasive ductal carcinoma of the pancreas is an independent factor for poor prognosis. This antibody recognizes a protein of 95 kDa, which is identified as villin. It is a major constituent in the microvilli, which compose the brush border of epithelial cells forming absorptive surfaces of the intestinal and renal proximal tubular epithelia. Anti-Villin labels the brush border area in the gastrointestinal mucosal epithelium and urogenital tract. Among neoplasms, villin is predominantly expressed in tumors of colorectal origin. Antibody to villin is useful in identifying malignant cells from primary and metastatic colorectal carcinomas. The protein encoded by this gene is a member of the transmembrane 4 superfamily, also known as the tetraspanin family. Most of these members are cell-surface proteins that are characterized by the presence of four hydrophobic domains. The proteins mediate signal transduction events that play a role in the regulation of cell development, activation, growth and motility. This encoded protein is found in the asymmetrical unit membrane (AUM) where it can complex with other transmembrane 4 superfamily proteins. It may play a role in normal bladder epithelial physiology, possibly in regulating membrane permeability of superficial umbrella cells or in stabilizing the apical membrane through AUM/cytoskeletal interactions. This MAb recognizes an intermediate filament protein of 46 kDa, identified as cytokeratin 20 (KRT20). KRT is abundantly expressed in goblet cells and enterocytes of the gastrointestinal tract. It has been detected in adenocarcinomas of the colon, stomach and biliary tract. The metal-responsive element (MRE)-binding transcription factor (MTF-1) stimulates the expression of metallothioneins in response to the exposure of cells to heavy metals. MTF-1 contains six zinc fingers in the DNA binding domain. The phosphorylation of MTF-1 in response to metal exposure appears to play a significant role in the ability of MTF-1 to activate metallothionein transcription. In addition to its role in metallothionein activation, MTF-1 is involved in a post-transcription regulatory complex for ribosomal protein S25.
